Description
💊 AMANTADINE 129 mg EXTENDED-RELEASE CAPSULE
Neurological Support with Sustained Relief
COMPOSITION:
-
Amantadine Hydrochloride 129 mg
(Extended-release capsule for once-daily use)
WHAT IS AMANTADINE 129 mg?
Amantadine 129 mg is a centrally acting dopaminergic and NMDA receptor antagonist used primarily in the treatment of Parkinson’s disease and drug-induced extrapyramidal symptoms. This extended-release formulation is designed to maintain steady blood levels over a 24-hour period, enhancing convenience and tolerability.
It is also used to manage fatigue in multiple sclerosis (MS) and has been explored for off-label uses in various neurological disorders.
MECHANISM OF ACTION:
Amantadine’s therapeutic effect is derived from a dual mechanism:
-
Dopaminergic Activity
-
Promotes release of dopamine and inhibits its reuptake in the brain.
-
Alleviates motor symptoms of Parkinsonism, including rigidity, tremor, and bradykinesia.
-
-
NMDA Receptor Antagonism
-
Blocks N-methyl-D-aspartate (NMDA) receptors, modulating excitatory glutamate transmission.
-
Helps reduce dyskinesia and prevent neurotoxicity from excessive stimulation.
-
INDICATIONS:
-
Parkinson’s Disease (PD) – monotherapy in early stages or adjunct in advanced PD
-
Drug-induced extrapyramidal symptoms (e.g., from antipsychotics)
-
Fatigue in multiple sclerosis (MS) (off-label use)
-
Post-viral fatigue syndromes (investigational use)
-
Traumatic brain injury-related fatigue (off-label)
DOSAGE & ADMINISTRATION:
-
Adults (extended-release formulation):
-
129 mg once daily
-
Dosage may be titrated based on clinical response and renal function
-
For some indications, higher total daily doses (e.g., 258 mg) may be used in divided or adjusted formulations
-
Capsules should be swallowed whole, not chewed or crushed.
Take at the same time each day, preferably in the morning to avoid insomnia.
KEY BENEFITS:
✅ Once-daily dosing for enhanced adherence
✅ Reduces motor symptoms and dyskinesia in Parkinson’s disease
✅ Improves fatigue and cognitive alertness in certain neurological conditions
✅ Offers an alternative to anticholinergics in drug-induced Parkinsonism
✅ Fewer peaks and troughs with extended-release formulation
